Linear Monofunctional PEG Derivatives

 

 

Linear monofunctional PEG derivatives have a reactive group at one end of the PEG and the other end is capped with a methoxy group or a sugar group.

 

Linear monofunctional PEG derivatives have the following general structure:

where X is the functional group or active group

and CAP can be either a methoxy group or a sugar molecule

JenKem Technology offers the following linear PEG NHS esters as regular products to meet with your different PEGylation requirements. Other PEG NHS esters with different molecular weights are available through custom synthesis.

 

GLUC-NHS-35K: Glucose PEG NHS Ester, MW 35000

 

The N-hydroxysuccinimide ester (or NHS ester) ester in this product will react with the amino group of lysine(s) or the N-terminal amine in biological active molecules to produce a stable amide linkage(s). The presence of glucose group will increase the selectivity of the PEGylation reaction. Glucose PEG NHS Ester will couple to the targeted molecules at pH 7-8. The PEGylation reaction can be finished in about 1 hour at ambient temperature. The NHS ester in this product is Succinimidyl Carboxymethyl Ester.

The average molecular weight of JenKem's
Glucose PEG NHS Ester is 35,000 Da. Substitution is 90% or greater. This product is basically free of active difunctional PEG impurities.

Methoxy PEG Carboxyl is the precursor to make Methoxy PEG Succinimidyl Carboxymethyl Ester (M-PEG-SCM). Methoxy PEG Carboxyl is much stable compared with M-PEG-SCM and can be used similarly to the corresponding NHS ester. M-PEG-SCM is chromatographically purified and is basically free of difunctional PEG impurities. The purity of M-COOH-PEG is 95% or greater.

The Nitrophenyl Carbonate group (NHS ester) in M-PEG-NPC will react with the amino group of lysine(s) or the N-terminal amine in biological active molecules to produce a urethane linkage. Urethane linker is also a stable linker but it is not as stable as amide linker. The reaction between M-PEG-NPC and amine containing molecules will release a p-nitrophenol which can be easily followed by UV spectroscopy.

M-PEG-MAL reacts with the thiol groups on cystein side chains in proteins and peptides to generate a stable 3-thiosuccinimidyl ether linkage. This reaction is very selective and can take place under mild conditions at about pH 5.0-6.5 in the presence of other functional groups.
